    题名: Role of mannose receptor (MR) in cockroach allergen-induced allergic inflammation

    摘要: RATIONALE: Early life exposure to cockroach allergen can lead to allergic sensitization, and an increased risk for asthma. A mouse model of cockroach allergen induced asthma has been experimentally well-established. We have recently demonstrated that mannose receptor (MR) mediates cockroach allergen recognition, uptake, and induced secretion of inflammatory cytokines.We aimed to test the hypothesis that MR may be critical in cockroach allergen induced airway inflammation and allergic responses.METHODS: Mouse models of cockroach allergen induced asthma were developed in C57BL/6 wild type (WT) and MR-deficient mice (MR-/-) using cockroach extract (CRE). Pulmonary inflammation, histology, serum antibody response, and cytokine expression profile were determined in these mice. Differential immune responses to CRE in splenocytes from MR-/- and WT mice were also examined. RESULTS: Significant recruitment of macrophages, eosinophils, neutrophils, and lymphocytes was observed in the BAL fluid of WT mice after exposure to CRE. Histological examination of the WT mouse lungs following CRE treatment showed intense peri-bronchial and peri-vascular infiltrates. Furthermore, serum levels of cockroach allergen specific IgE(sIgE), IgG1, and IgG2 and lung tissue IL13, IL-6, IL-1a, TNF-a, and MR expression were significantly increased in CRE-challenged WT mice. In contrast, MR-/- mice were significantly protected against most of these major asthmatic phenotypic alterations induced by CRE, including eosinophil recruitment, peri-bronchial and peri-vascular infiltrates, sIgE, and cytokine expression. Additionally, IFN-gamma was significantly increased in CRE-treated splenocytes from MR-/- compared to those from WT mice. CONCLUSIONS: These findings suggest a role for MR in CRE-induced pulmonary allergic inflammation in mouse models.
